First of all, we talk about which incubators are suitable for laboratory use. The cultured cells in vitro need to be maintained at a constant temperature like the cells in the body. Generally, the optimum growth temperature is 37°C, and the temperature difference should not exceed ±0.5°C. When the temperature rises by 2°C, it becomes unfavorable to the survival of the cells, and the temperature will reach above 40°C and the cells will soon die. Therefore, an incubator or CO2 incubator with precise temperature control is the best choice for cell culture in the laboratory. (1) Thermostatic incubator: It should be selected from water-type or transistor-type self-controlled temperature incubator. Such incubators have high sensitivity and stable temperature control. The main heating methods can be divided into two types: water jacket type and electric film type. The electric heating film is directly heated by heating the electric film attached to the inner wall, and the water jacket type is heating the outer water layer to heat the interior again. Both heating methods have their own advantages. The electric heating film is heated more quickly and can be cultured within a short time so that the internal temperature reaches the desired temperature. Water jackets are better than stability. The thermostatic incubator is generally cheaper, and its disadvantage is that it is only suitable for closed culture.
(2) CO2 incubator: At present, carbon dioxide incubators have been widely used in most cell culture chambers. The advantage of the CO2 incubator is that it can provide a certain amount of CO2 (usually 5%) required for cell culture, and it is easy to stabilize the pH of the culture solution. It is suitable for open or semi-open culture. When using a culture bottle, the bottle cap can be loosened slightly to keep the inside of the culture bottle ventilated. To avoid contamination of the cells, the air in the incubator must be kept clean and ultraviolet radiation must be used regularly when using this culture method. Or alcohol disinfection, and in the incubator should be placed in a tank filled with sterile distilled water to prevent evaporation of the culture solution, maintaining the relative humidity within the box at 100%.
